Instructions for Microbiology Plate Pouring Kit 

 

1.      Remove number of petri dishes required for pouring. Leave remaining petri dishes in plastic sleeve and re-seal. Each 250 ml bottle of media provides approximately 10-15 plates.

 

  1. Place one bottle of media in a heating device to melt. There are two recommended heating options:

 

Microwave- Unscrew cap and leave resting atop bottleneck. Place in microwave and set for medium heat for approximately two minutes. Heat an additional 30 seconds at medium heat. Repeat this process until media in all liquid. Caution: Liquid media will be very hot. Wear appropriate gloves and eye protection to handle bottle. Avoid shaking or swirling media to prevent boil over.

 

Hot water bath: Unscrew cap and leave resting atop bottleneck. Place bottle in boiling water. Media is ready to pour when liquefied. Caution: Liquid media will be very hot. Wear appropriate gloves and eye protection to handle bottle. Avoid shaking or swirling media to prevent boil over

 

  1. Lift lid of dish enough to pour media into petri dish. Fill to a level of approximately 3/16” or 4mm.  

 

  1. Place lid on filled dish bottom and allow to cool. Approximately 30-40 minutes. Place cooled prepared plates in a refrigerator until ready to use.

 

Note: Used media plates should to decontaminated after use. Bleach can be poured over each plate. Place decontaminated plates in a plastic bag and seal. Dispose of plates according to your institution’s policy and procedures.
